i was just wondering if i can test my pres for their true voltage so i can correctly set my amp, just touching positive and negative on the multimeter gives no readout

its a panasonic with a claimed 5v preouts and a kicker kx400.1 which accepts up to 5v should i turn the gain all the way counterclockwise or clockwise?
